IntroductionColorectal cancer is a common malignant tumor in the world. The incidence of colorectal cancer shows a rising trendency. Accumulated evidence has established that carcinogenesis is associated with alterations in cellular oncogenes and in tumor suppressor genes necessary for malignant transformation. About twenty percent of colorectal cancer is characterized by a familial aggregation. Hereditar-y non ? polyposis colorectal cancer (HNPCC) is thought to be the most common form of inherited colorectal cancer. HNPCC is an autosomal dominant disorder which accounts for 5% ? 15% of colorectal cancer cases. Most cases of HNPCC and part of sporadic colorectal cancer are caused by a defect in the DNA mismatch repair gene ( MMR) but not in oncogenes or tumor suppressor genes. Tumors which have defective mismatch repair gene show a characteristic molecular phenotype, termed microsatellite instability ( MSI).Microsatellites are polymorphic, short, tandem repeat segments dispersed throughout the human genome. MSI is a form of genetic instability characterized by expansions and contractions of simple sequence repeats in DNA. It occurs because of a defect in DNA MMR. Nowadays, MSI has been paid more attention to because of its key role in the initiation and development of carcinoma. MSI is considered a new mechanism of colorectal cancer.In our experiment, polymerase chain reaction ? single strand conformation polymorphism (PCR ? SSCP) and sliver ? staining methods were performed to detect incidence of MSI in 29 cases of CRC with familial predisposition and in 30 sporadic CRC. The goal is to discuss the molecular mechanisms of CRC with familial predisposition and the possibility to discover disease in early stage, directclinical treatment and judge the prognosis.Materials and MethodsI. Materials1. Samples All samples used were obtained from paraffin ? embedded tissues of the surgical resected specimens of patients admitted to the 1 st Affiliated Hospital of China Medical University during 1997 -2003.2. Reagents Primer; BAT25, BAT26, D5S346, D17S250 and D2S123. (GIAGEN company); TaKaRa Taq?, Proteinase K, Agarose Regular, DNA Marker DL2000 ( TaKaRa Biotechnology company ) ; the other reagents made in China were all analytical pure.II. MethodsSerial five 8 ? jxm thick sections of selected tissue blocks were obtained. Corresponding samples of colorectal mucosa that were free of tumor and metaplasia , were obtained from each case as control tissue. Sections were dewaxed. Then genomic DNA was purified by proteinase K digestion and phenol ? chloroform extraction. The ratio of A260 and A280 was measured by ultraviolet spec-troscopic photometer. The concentration of DNA was calculated. PCRs were performed to amplify colorectal lesions and corresponding normal DNA. After amplification, PCR products were separated by 2% Agarose gels to detect DNA. Then PCR products were electrophoresed on non ? denaturing 8% ( 29 :1) poly-acrylamide gels, and bands were visualized by silver staining.Alterations in the microsatellites in the form of changes in the size of DNA strands were detected by comparing tumor DNA and normal DNA. It was judged negative if their strands have no difference. MSI was defined as a band shift in either of the two alleles or the appearance of a band with different sizes in the tumor or normal sample. Using a panel of five microsatellites, a specimen was defined as high ? level MSI ( MSI ? H) if two or more of the five markers displayed instability,as low -level (MSI ? L) if one marker displayed instability, and as microsatellite stable (MSS) if none of the markers exhibited MSI.III. Statistical analysisStatistical analysis was performed using the t ? test and Fishers exact test. P 0. 05 ).MSI incidence rates in different reports vary. We therefore postulate that MSI rates may be associated with markers, the criteria for definition of MSI, the number of samples, race and different areas.Our results demonstrate that MSI plays an important role in CRC with familial predisposition. MSI analysis is very useful in detecting tumors with mismatch repair genes as the first step. The verticle relatives of MSI carriers have a higher risk of HNPCC. We expect to discover HNPCC in early stage among those people by surveillance and have the possibility of early surgical intervention that could be curative.

Culinary Union suggests Bill of Rights for MMA fighters
The Culinary Union, the biggest union representing workers in Nevada, stopped by the Nevada Athletic Commission's meeting on Wednesday to discuss ways to improve treatment for fighters. They want to see the NAC lead the combat sports world by pushing for reforms in MMA.
1. Equal protections for all fighters. Boxers are protected under the Muhammad Ali Act, which enacts certain rules over boxers that keeps them from getting exploited. Mixed martial artists are not.
2. Right to work. This would allow fighters to sign non-exclusive contracts and would prohibit contracts from automatically renewing. Champions' clauses, which keep champs attached to their contract as long as they hold the belt, would be a thing of the past.
3. Inalienable right to your own name, likeness and image. When Jon Fitch objected to handing over his image in perpetuity to the UFC for its video game, he was cut from the UFC and Dana White threatened to not do business with any fighters from Fitch's gym, American Kickboxing Academy. After Fitch played ball, he was brought back into the fold. This right would allow fighters to exercise the right Fitch wanted to.
4. Free market of sponsorships. This would not just allow fighters to get whoever they would like to sponsor them, but would also let them say no to their promotion's sponsor. In other words, Brock Lesnar could chug a Coors and Carlos Condit could have walked away from a Harley-Davidson if he wanted to pursue a sponsorship with Honda.
5. Transparency of contracts and payments. Fighters would get detailed financial statements from any event they participated in. This would be particularly important to fighters whose contracts earn percentages of pay-per-views or gates.
6. Fair share of revenues. In the NBA and NFL, athletes went through a lockout to fight for roughly half the league's revenues. Because Zuffa is a private company, their revenues are not public, so we have no idea how much revenue the fighters earn. This right would ensure fighters get at least a quarter of revenues.
7. Freedom of association. Fighters would be allowed to unionize in any way they see fit.
8. Right to healthcare insurance for training and fighting. Zuffa fighters are given medical coverage for both fight camps and fights, but this right would ensure it continued.
9. Right to fair fights. The UFC's matchmaking system generally provides fair and evenly matched bouts, but things get murky on subjects like who deserves a title fight. Timing often decides title matches as much as an independent ranking system. This right would call for a transparent ranking of fighters.
10. Professionalism. From the union: "You have the right to be treated with common courtesy and professional respect by other fighters and by promoters and managers. For mixed martial arts to become a mainstream sport accepted by the general public, participants in the sport must act in accordance with commonly accepted standards of courtesy, decency and respect in their public interactions with one another and in their interactions with the public."
In other words, promotions couldn't fire one fighter because of a tweet about rape while not firing another for jokes about child molestation. UFC executives would probably have to stop dropping F-bombs at Twitter followers.

Latino voters: Missed opportunity for Republicans?

With the Republican contenders meeting in Mesa, Ariz., Wednesday night for their final debate before Tuesday?s primaries in that state and in Michigan, the issue of illegal immigration will likely get another turn in the spotlight and the GOP will get another reminder of the general election difficulties it faces with Hispanic voters.
None of the four remaining GOP contenders has voiced support for a broad amnesty that would allow younger illegal immigrants to become permanent legal residents.

Of the four, former House Speaker Newt Gingrich has the most accommodating policy toward illegal immigration, calling for local citizen review boards to allow some long-term illegal residents to remain in the United States.
While immigration hasn?t been a dominant issue in this GOP presidential contest, it has a deep impact in Arizona where Republican Gov. Jan Brewer, signed SB 1070, a crackdown on illegal immigrants in 2010.?Brewer has not yet endorsed a Republican presidential contender.
President Barack Obama?s Justice Department is trying to overturn that Arizona law and the Supreme Court hears oral arguments in that case on April 25.
The GOP contenders are unlikely to directly criticize the Arizona law in Wednesday's debate on Brewer?s home turf. More than two-thirds of likely Republican primary voters in Arizona said they?d be more inclined to vote for a presidential candidate who backs SB 1070, according to the NBC News/Marist Poll released Wednesday.
The GOP contenders walk a fine line: hard-hitting rhetoric on immigration is popular with conservative primary voters, but may be costly in the fall because Latinos seem likely to account for a bigger share of the general electorate in battleground states like Colorado and Nevada than they did four years ago.
A new NBC News poll shows that GOP presidential hopefuls Mitt Romney and Rick Santorum are neck-and-neck in Michigan, but Romney has a comfortable lead in Arizona. NBC's Chuck Todd reports.
With Brewer now one face of the party on the issue, the GOP has come a long way from 2004 when President George W. Bush -- who said ?family values don?t stop at the Rio Grande? -- won an estimated 43 percent of Latino voters. Republican candidate John McCain won an estimated 31 percent of Latino voters in 2008.
In the 2008 election, Arizona went for its own senator, McCain. This year, its 11 electoral votes are an alluring target for Obama?s strategists. But the Democrats? ?chances of it flipping are pretty minimal? this year due to the conservatism of white voters there, said Ruy Teixeira, a political demographer and senior fellow at the Center for American Progress, a Democratic-allied think tank.
In the NBC News/Marist Poll of Arizona voters, in a hypothetical contest between Obama and Mitt Romney, 45 percent said they?d support Romney and 40 percent said they?d back Obama.
But overall in the general election, ?The Latino vote is going to be absolutely crucial in 2012,? Teixeira said at a recent conference on Latino voters at American University in Washington.?
In Nevada, for example, Teixeira?projects a four percentage-point increase in the minority share of the vote and a five-point decline in?white working-class voters? share of the vote.
The NOW panel expects immigration to be a hot-button issue during Wednesday night's GOP presidential debate in Arizona, but will the issue rise to the forefront of national attention?
If Obama can win 80 percent of minority voters nationally, ?he could get shellacked? among white voters ?as badly as?Democratic congressional candidates were in 2010, when they lost the white working class by 30 points? and yet ?he could almost survive that level of shellacking,? Teixeira argued.
For every presidential hopeful, a $1 million donor
Even in Pennsylvania, where Latinos were only four percent of the 2008 electorate, they may end up being crucial, Teixeira said.
He predicted that Obama will lose?among Pennsylvania?s white working-class voters, but ?all he has to do is not get totally wiped out. He can afford a 15-point loss, he can afford a?20-point loss, what he doesn?t want is 30-point loss? among white working-class voters.
?If he can get the Latino vote mobilized and motivated to vote for him at a high level, I think it very much reinforces his chances of taking the state,? he said.
Polling of registered Latino voters for Univision last month suggested that GOP opposition to the DREAM Act will make it impossible for most Latinos to vote Republican in November. In the Univision poll, 85 percent of Latino voters supported the DREAM Act.
Passed by the House, but rejected by the Senate in 2010, the DREAM Act would allow non-citizens under age 30 who entered the United States illegally before their 16th birthday to remain as legal U.S. residents, as long as they?d committed no serious crimes, earned a high school diploma, or served in the military.
Even though more than 60 percent of Latinos are U.S. born, and thus American citizens, most Latino registered voters also say they know someone -- sometimes a family member -- who is an illegal immigrant. That personal connection is one reason why the DREAM Act has become a litmus test.
All but three GOP senators voted against allowing a vote on the DREAM Act in 2010; all but five Democratic senators voted for it.
Obama?s support for the DREAM Act may be an electoral liability in states such as North Carolina -- Democratic Sen. Kay Hagan and three of the state?s Democratic House members voted against it in 2010 -- and in Indiana: Democratic Senate candidate Rep. Joe Donnelly voted against it. Obama carried both North Carolina and Indiana in 2008.
But elsewhere Obama?s support for the bill seems likely to help him win Latino votes.
?A lot of Latinos are very upset about Obama?s deportation policies? -- there were a record number of deportations in 2010 -- ?there?s a disappointment there,? Teixeira said, but he contends that what he calls the ?anti-immigrant tenor? of the Republican Party ?is pushing Latinos into the arms of the Democratic Party.?
Stanford University political scientist Gary Segura said that the GOP ?has missed a strategic opportunity? to win over Latinos. Obama, he said, ?is assuming he has support (among Latinos) that he may not have, but he might ultimately get away with it anyway ... because of Republican messaging on the (immigration) issue.?
Republican National Committee spokeswoman Alexandra Franceschi disputed the idea that immigration will dominate Latino voters? decisions in November.
Romney's organization meets Santorum's momentum
Polls, she said, ?show that the number one issue that Hispanics across the country are considering when they?re going to make an electoral decision is jobs and the economy. I think the immigration issue kind of gets blown out of proportion ... .?

The RNC launched a Latino outreach program last month, hiring Bettina Inclan, a former strategist for Florida Gov. Rick Scott, to direct that effort which will put RNC field workers in Florida, New Mexico, Colorado and Nevada to mobilize Latino Republicans.
The tension over immigration in the GOP has been between Gingrich and Romney.
In a debate last December, Gingrich referred to ?someone who's been here 25 years, somebody who has been a good local citizen, may well belong to your church, has children and grandchildren in the United States,? and argued that ?I do not believe the people of the United States are going to send the police in to rip that kind of person out and ship them out of this country ... .?
But he also said that most illegal workers in the United States ?should go home immediately? and ?we should make deportation dramatically easier.?
Gingrich in one ad branded Romney as ?the most anti-immigrant candidate,? and despite taking down that ad, stood by that label when asked about it in a debate last month.
Romney has said, ?I am pro-immigrant. I want people to come to America with skill and vitality and vibrancy. I want them to come legally.?
He has also said, ?I'm not going around and rounding people up and deporting them.? He proposes that legal immigrants receive a work permit. ?People who do not come here legally do not get a work permit. Those who don't get work will tend, over time, to self-deport,? he said.
Romney said last month that he?d veto the DREAM Act, but he supports a version of it that would open a path to citizenship to those who serve in the U.S. military.
Despite the general Latino support for the DREAM Act, Romney did win 54 percent of Latino voters in Florida?s Jan. 31 Republican primary.
Former Pennsylvania senator Rick Santorum voted against the 2006 comprehensive immigration reform bill which included a version of the DREAM Act.
He supports legal immigration, in order to boost population growth -- ?We are not replacing ourselves,? he warned last month -- and because ?immigrants bring a vitality and a love of this country that infuses this country with great energy.?
But he said, ?people who have come to this country illegally have broken the law repeatedly? by working here and must be deported.
The maverick GOP contender, Texas Rep. Ron Paul, who voted against the Dream Act in 2010, said in a debate last month, ?We spend way too much time worrying about the border between Afghanistan and Pakistan. Use some of those resources on our own border.?
But Paul also seemed to imply he?d tolerate some illegal immigration in a boom economy: ?The weaker the economy, the more resentment there is when illegals come in. If you have a healthy, vibrant economy, it's not a problem; we're usually looking for workers.?

OLYMPIA, Wash. (AP) ? Josh Powell had hundreds of images of disturbing cartoon sex and graphic depictions of incest on his home computer, something Utah authorities investigating his wife's disappearance kept private for about two years, according to files released Friday.
A Washington state psychologist who had previously said Powell's strong parenting skills should allow him to continue supervised visits altered his assessment after reviewing the images, calling them a "great concern" and suggesting a more intense evaluation of the father. That conclusion came just six days before he killed himself and two sons in an explosive house fire two weeks ago.
Many of the approximately 400 images described in the state social services included sexual depictions of popular cartoons, including child-focused characters such as Rugrats, Dennis the Menace and SpongeBob SquarePants. Another 15 images showed 3-D depictions of sex involving parents and their children.
Powell had initially told the psychologist that he visited only pornography sites featuring adults.
"Given the gaps of information about Mr. Powell there seems reason to conclude he may not presently be a stable and appropriate resource for his children," wrote Dr. James Manley in a follow-up report.
Washington officials overseeing the care of Powell's children first got word of the graphic content in November, when a Utah investigator wrote in an email that they were looking to disclose to social workers about 5-6 images recovered in 2009 shortly after Susan Powell's disappearance.
But two months later, social worker Forest Jacobson wrote that the images weren't received until mid-January and told the psychologist "it is now clear there are many more images than initially indicated."
It wasn't clear why it took so long for authorities to reveal the information. A spokesman for the West Valley City police in Utah declined comment.
In other documents released under Washington public records laws, social workers detailed how they were concerned that Josh Powell would speak disparagingly of his in-laws, Chuck and Judy Cox, in front of his children.
The documents also state that Charlie Powell was beginning to remember about his mother, and that he said "he remembers she left because grandpa was mean to her" ? apparently a reference to Susan Powell's father. Caseworkers believed that Charlie was repeating what his father had been saying.
In interviews with mental health professionals, Powell said he feared a "militant faction" of the Mormon church might kidnap his two sons. Braden and Charlie were overheard saying that "Mormons" were "trying to steal" them and "harm" them. When Charlie told his father that they were "good Mormon boys," Powell abruptly responded that they were not Mormon and not to let anyone tell them they are.
Charlie once told school personnel how to bury an animal so it wouldn't be discovered. Personnel only noted this because of their mother's disappearance.
Powell was a suspect in Susan Powell's 2009 disappearance from their home in West Valley City, Utah. He had always claimed that he didn't know what happened to his wife. He took the boys ? then 2 and 4 ? on a midnight camping trip in freezing weather in the Utah desert, he said, and when he returned home the next day authorities were at the house looking for her.
Weeks later, he moved the boys to the home of his father, Steve Powell, in Puyallup. After Steve Powell's arrest on voyeurism and child pornography charges last fall, the boys were removed from the house and turned over to Susan Powell's parents.
A social worker brought them to Josh Powell's rental home for what was supposed to be a court-sanctioned supervised visit. Josh Powell let the boys inside, locked the social worker out, hit them with a hatchet and set fire to gasoline, authorities said.
The records indicate that Susan Powell remained a focal point in the family. Charlie talked about how her favorite color was purple. Powell talked about how she made tasty pumpkin pies from pumpkins that they had grown themselves.
As they ate pancakes, bacon and orange juice one day in early November, Braden looked at Powell and said: "I wish mommy was here." Powell said he did, too.
The next month, as they decorated for Christmas, Charlie showed the visit supervisor a stocking with "SUSAN" lettered across the top. He later found his mother's favorite stained glass ornament, and Powell said he could hang it on the tree.
In notes from Jan. 29 ? the day of the last visit before Josh Powell killed his children ? the social worker wrote that the boys made Shrinky Dink toys with him. When the visit was over, Josh Powell walked Charlie and Braden out to the car, buckled them into their booster seats, told them he loved them and said he would see them as soon as he could.
"Be happy and have a good time," he said.
